I would recommend doing a search log analysis using the search logs of a Magento shop and do a full analysis such as is explained here:
http://faculty.ist.psu.edu/jjansen/academic/jansen_search_log_analysis.pdf

This will probably yield you to see that high frequency queries have to be tuned manually to avoid false-positives and other errors that can occur with mass automation of error-tolerance.  Then for the bulk of queries the best is to automate.  If you are trying to automate for a single language (ie. English) Soundex might help, if not or if you really want to achieve the best coverage, edit distance algorithms are the best tool.  They cost a lot in terms of hardware unless you can bring your catalog into an in-memory indexer of some sort.  Even yet, doing fast autosuggest like Google might be time consuming unless you use something like:  http://commerce.exorbyte.com which is built to do all that.
